Re-endothelialization of BioVaM using porcine derived endothelial cells

چکیده
Introduction A persistent problem of the generation of 3D tissues for therapeutic application is efficient nutrient supply and removal of waste products within complex constructs. Depending on the tissue to be reverse engineered, a more or less intense supply of substrates and oxygen must be provided, as diffusion processes are limiting as tissue thickness increases. Thus, the use of decellularized, biological vascularized matrices (BioVaM) with preserved arterial and venous pedicles, aid the generation of perfusable tissue constructs in vitro. To prevent vessel occlusion when implanted, a functional endothelium must line the graft vessel bed. Thus, we describe the re-endothelialization of BioVaM with porcinederived endothelial cells (EC) in vitro.

Cilostazol Activates Function of Bone Marrow-Derived Endothelial Progenitor Cell for Re-endothelialization in a Carotid Balloon Injury Model
BACKGROUND Cilostazol(CLZ) has been used as a vasodilating anti-platelet drug clinically and demonstrated to inhibit proliferation of smooth muscle cells and effect on endothelial cells. However, the effect of CLZ on re-endothelialization including bone marrow (BM)-derived endothelial progenitor cell (EPC) contribution is unclear. We have investigated the hypothesis that CLZ might accelerate re...
متن کاملIn vitro endothelial wound repair. Interaction of cell migration and proliferation.
Most re-endothelialization requires both cell migration and cell proliferation. To study the association between cell migration and the initiation of DNA synthesis in an in vitro wound model, confluent cultures of porcine aortic endothelial cells grown on glass coverslips were scraped to remove half of the monolayer. Treatment with 1 ng/ml of taxol, a microtubule stabilizing drug, for 24 hours ...
متن کاملCombination coating of chitosan and anti-CD34 antibody applied on sirolimus-eluting stents can promote endothelialization while reducing neointimal formation
BACKGROUND Circulating endothelial progenitor cells (EPCs) capture technology improves endothelialization rates of sirolimus-eluting stents (SES), but the problem of delayed re-endothelialization, as well as endothelial dysfunction, has still not been overcome. Therefore, we investigated whether the combination coating of hyaluronan-chitosan (HC) and anti-CD34 antibody applied on an SES (HCASES...
متن کاملInhibition of miR-92a improves re-endothelialization and prevents neointima formation following vascular injury
AIMS MicroRNA (miR)-92a is an important regulator of endothelial proliferation and angiogenesis after ischaemia, but the effects of miR-92a on re-endothelialization and neointimal lesion formation after vascular injury remain elusive. We tested the effects of lowering miR-92a levels using specific locked nucleic acid (LNA)-based antimiRs as well as endothelial-specific knock out of miR-92a on r...
متن کاملMagnetic forces enable rapid endothelialization of synthetic vascular grafts.
BACKGROUND Synthetic vascular grafts cannot be used in small vessels because of graft failure caused by thrombosis and neointima formation. Rapid endothelialization may overcome this limitation. We hypothesized that a magnetic graft would be able to capture and retain endothelial cells labeled with paramagnetic particles. METHODS AND RESULTS Porcine blood derived endothelial cells were allowe...
